For most substantial agency work, yes. The tender documents will name the applicable register or scheme and the required category, which typically caps the contract value you can be awarded. Assessment requires audited financials and takes weeks, so complete it before a tender you want appears rather than during.
On larger contracts they are scored criteria with contractual reporting obligations attached. Apprentice ratios, local employment, and spend with social enterprises, Aboriginal businesses and disability enterprises all feature. Because you will be held to what you promise, cost the commitments properly and only offer what you can actually deliver.
